1A: COB Test :
Requirements (Instruments, Chemicals & Other) :

1. Milk sample
   
2. Test tubes
   
3. Water bath
   

Procedure :

1. Take 5ml of milk from each sample in a separate test tube.
   
2. Place the test tubes in a boiling water bath for 5 minutes.
   
3. Remove the tube from water bath without shaking.
   
4. Observation is No clot formation- Negative and Clot formation-Positive
   

1B: MBRT Test :
Requirements (Instruments, Chemicals & Other) :

1. Milk sample
   
2. Standard methylene blue solution
   
3. Test tubes
   
4. Water bath
   

Procedure :

1. Take 10 ml of milk from each sample and pour in a two separate test tube
   
2. Add 1 ml of standard methylene blue solution in both the test tubes.
   
3. Invert the test tube to mix the milk and methylene blue solution.
   
4. Place the test tube in a thermostatically water bath maintained at 37°C.
   
5. Observe the test tubes after 30 min for decolourization /reduction of dye.
   
6. Place the test tubes again in the water bath for 2 hr if not decolorizes
   
7. Observe the test tubes after 2 hr for decolourization /reduction of dye
   
8. Repeat the procedure of incubation for 5 hr if not decolorizes
